Design: The experimental procedure was performed according to the standard protocol provided by Pacbio, including the processes of sample quality testing, library construction, library quality testing and library sequencing, and library construction included the following steps: 1. Use g-TUBE to interrupt the DNA sample. 2; 2. damage repair of the interrupted DNA sample; 3. performing end repair on the DNA; 4. connect the dumbbell junctions; 5. perform exonuclease digestion. 6; 6. use BluePippin to screen the target fragments and obtain the sequencing library.

Descurainia sophia Willd. belongs to the Descurainia of Cruciferae. Its dried mature seeds are called Nantinglizi, which is a traditional Chinese medicine commonly used for purging the lung, relieving dyspnea, and alleviating edema in clinical practice, and has the significant pharmacological efficacy for cardiovascular disease, hyperlipidemia, etc. The seeds of D. sophia are rich in unsaturated fatty acids, glucosinolates, sterol, and have a variety of biological activity compounds. We assembled a high-quality chromosome-level reference genome of Descurainia sophia genome, using PacBio long reads in combination with Illumina short reads genome sequencing and HiC technology. The high-resolution reference genome generated by this study will provide accurate sequence information for glucosinolate biosynthesis research of D. sophia.
